Monocyte TREM-1 Levels Associate With Anti-TNF Responsiveness in IBD Through Autophagy and Fc?-Receptor Signaling Pathways

چکیده
The expression of Triggering Receptor Expressed on Myeloid cells ( TREM)-1 has been described as a predictive marker for anti-Tumor Necrosis Factor (TNF)-? monoclonal antibody (mAb) therapy responsiveness in patients with inflammatory bowel disease (IBD). Here we investigated TREM-1 specifically CD14+ monocytes relation to anti-TNF response. pretreatment levels Crohn’s (CD) were outcome mAb therapy, low associated response anti-TNF. FACSorting different showed that differentiation towards regulatory CD206+ M2 type macrophages by was suppressed high expression. Activity the Fc?-Receptor and autophagy pathway, both necessary anti-TNF, decreased TREM-1. We confirmed activity pathway CD did not respond it negatively correlated patient cohort. In conclusion, our results indicate associate Fc?R resulting upon treatment, which may explain non-response IBD .

TREM-1 SNP rs2234246 regulates TREM-1 protein and mRNA levels and is associated with plasma levels of L-selectin
High levels of TREM-1 are associated with cardiovascular and inflammatory diseases risks and the most recent studies have showed that TREM-1 deletion or blockade is associated with up to 60% reduction of the development of atherosclerosis. So far, it is unknown whether the levels of TREM-1 protein are genetically regulated. Moreover, TREM family receptors have been suggested to regulate the cel...
متن کاملMonocyte TREM-1 membrane expression in non-infectious inflammation
Sterile or non-infectious inflammation and severe infection are accompanied by very similar events. Damage (or danger)-associated molecular patterns and pathogen-associated molecular patterns trigger the release of inflammatory mediators and modulate the expression of cell surface receptors. The increased expression on monocytes and neutrophils of triggering receptor expressed on myeloid cells ...
متن کاملCB2 cannabinoid receptor agonist JWH-015 modulates human monocyte migration through defined intracellular signaling pathways.
Recruitment of leukocytes to inflammatory sites is crucial in the pathogenesis of chronic inflammatory diseases. The aim of this study was to investigate if activation of CB2 cannabinoid receptors would modulate the chemotactic response of human monocytes. Human monocytes treated with the CB2 agonist JWH-015 for 12-18 h showed significantly reduced migration to chemokines CCL2 and CCL3, associa...
متن کاملHigh CerS5 expression levels associate with reduced patient survival and transition from apoptotic to autophagy signalling pathways in colorectal cancer
Ceramide synthase 5 is involved in the de novo synthesis of ceramide, a sphingolipid involved in cell death and proliferation. In this study, we investigated the role of ceramide synthase 5 in colorectal cancer by examining ceramide synthase 5 expression, clinico-pathological parameters and association with survival/death signalling pathways in cancer. Immunohistochemical analysis of CerS5 was ...
متن کاملBenefit of Earlier Anti-TNF Treatment on IBD Disease Complications?
BACKGROUND Anti-tumour necrosis factor [anti-TNF] treatment was demonstrated to have disease-modifying abilities in inflammatory bowel disease [IBD]. In this study, we aimed to determine the effect of anti-TNF treatment timing on IBD disease complications and mucosal healing [MH]. METHODS The following IBD-related complications were tested in relation to timing of anti-TNF therapy start in ne...
